> May Grunwald-Giemsa staining | > Giemsa staining | > Rapid staining (panoptic)

 May Grunwald-Giemsa staining
  • Cover the slide with 10 drops of pure May-Grunwald stain and allow it to react for 3 minutes.
  • Add drops of neutralized water to the slide and agitate gently to homogenize. 
    Allow it to react for 1 minute.
  • Pour off the mixture. Do not wash up. 
  • Cover the slide with a diluted Giemsa stain prepared extemporaneously with neutralized water:
       - Giemsa stain diluted at 1/10 for 10 minutes
       - Giemsa stain diluted at 1/20 for 20 minutes for better contrast 
  • Wash up with neutralized water. 
  • Wipe the slide on the reverse side of the smear.
  • Dry: drying oven, hair drier, electric fan.  

ATTENTION
THE MAY-GRUNWALD STAIN IS NOT STABLE FOR IT CONTAINS ALCOHOL
IT IS IMPORTANT TO FIX THE SPECIMEN PRIOR TO STAINING  

 Giemsa staining
  • Fix with methanol.
  • Cover the slide with a 1/20 dilution of Giemsa stain prepared extemporaneously with neutralized water. Allow it to react for 20 to 30 minutes (experience will determine the optimal time).
  • Wash up with neutralized water.
  • Wipe the slide on the reverse side of the smear.
  • Dry: drying oven, hair drier, electric fan  
 Rapid staining (panoptic)
  • Cover the slide with 10 drops of panoptic stain and allow to react for 90 seconds.
  • Add the same quantity of hematology adjuvant and allow to react for 90 seconds.
  • Rinse with the same adjuvant.
  • Wipe the slide on the reverse side of the smear.
  • Dry the slide immediately, paying careful attention, for the smear is fragile.
